dc.description.abstract | Fruktozila peptīdu oksidāzes, pazīstamas arī kā Amadoriāzes, ir enzīmu saime, kas katalizē fruktozilaminoskābju deglikozilāciju. Šī darba mērķis ir novērtēt termālo stabilitāti un enzimātisko aktivitāti Amadoriāzes enzīma mutantiem, C16 un D02, kurus izstrādāja ārvalstu kolēģi, izmantojot skaitļošanas metodes. Darba gaitā tika veikta veikta ekspresija E. coli šūnās un proteīnu attīrīšana, izmantojot šķīdumu hromatogrāfiju. Tika veikta proteīnu analīze, izmantojot biofizikālu un bioķīmisku analīžu metodes - termiskās nobīdes, cirkulārā dihroisma un enzimātiskās aktivitātes testus. Rezultāti uzrādīja termostabilitātes un viršas punktu palielināšanos abiem mutantiem, enzimātiskās aktivitātes un α-spirāles satura palielinošānos D02 variantam, bet ne C16 variantam, salīdzinot ar savvaļas proteīnu. | |
dc.description.abstract | Fructosyl amino acid oxidase (FAOx), also known as Amadoriase, are a family of enzymes that catalyze the deglycosylation of fructosyl amino acids. The aim of this work is to assess the thermal stability and the enzymatic activity of two Amadoriase mutants, C16 and D02, which have been designed via computational methods by external collaborators. For this work, protein expression in bacterial (E. coli) cells and protein purification using liquid chromatography techniques were done. Then, biophysical and biochemical analyses of the protein using thermal shift assay, circular dichroism and enzymatic activity assay were performed. Results showed an increase in thermal stability and melting points for both mutants and an increase in enzymatic activity and helix content for D02 and not for C16 relative to the wild type protein. | |
